Disclosed is an isolated naturally-occurring mutant human B-Raf polypeptide comprising one or more amino acid mutations, wherein the single amino acid mutations are located C-terminal to amino acid 300 in B-Raf, and wherein the amino acid mutations occur at one or more of positions 463, 465, 468, 585, 594, 595, 596 and 599 in B-Raf. Also disclosed are complements of nucleic acids encoding said mutant human B-Raf polypeptide, ligands binding selectively to said polypeptide and methods of detecting said mutations. Mutations in the sites disclosed are shown to have an oncogenic effects and inhibitory effects on kinases. |
