abstract | Methods of screening for attenuated strains of Mycobacterium tuberculosis complex, which comprise at least one inactive gene, are described. The screening comprises steps of isolating a Mycobacterium species to be screened, growing the isolated mutants for 5-10 weeks in a complete medium, transferring the mutants to minimal media and selecting the mutants based on their inability to grow in minimal media. The figure shows a Southern blot hybridization after EcoR1 digestion and probing with neo of eight recombinants that were unable to grow in a minimal medium (lanes 1-5 and 7-9) and one recombinant that failed the screen and grew in the minimal medium (lane 6).
