abstract |
A method for separating a protein molecule or a population of protein or peptide molecules from high molecular weight molecules or structures comprises contacting cells comprising the protein with at least one pore-containing matrix. The matrix retards the flow of high molecular weight molecules or structures, but does not retard the flow of soluble protein and peptide molecules and comprises one or more lysis/disruption/permeabilization compositions or compounds in an amount sufficient to lyse, disrupt or permeablise the cells. The matrix is then subjected to conditions that promote the flow of material through the matrix, wherein the conditions are sufficient to allow the proteins to pass through the matrix which the HMW molecules are trapped or bound to the matrix, thereby separating the protein and HMW molecules. Also described is an apparatus for extracting and isolating protein or peptide molecules comprising a housing containing the pore containing matrices and composition selected from chromatographic resins. |
