abstract Capsular polysaccharide can be produced from a pneumococcus by obtaining an opaque variant of the pneumococcus and maintaining the variant in a growth medium in contact with a gas having n oxygen concentration less than about 16% and/or a carbon dioxide concentration greater than 5% and then optionally isolating the capsular polysaccharide. A gas having super atmospheric concentration of oxygen can be use din the preparation of a gaseous medicament for the treatment of a pneumococcal infection. Also described are methods of making immunogenic preparation, and methods of assessing whether a test compound is useful in alleviating a pneumococcal infection.
