abstract |
A monoclonal antibody which specifically binds to a conserved epitope of prion proteins, designated as Gln-Tyr- Gln-Arg-Glu-Ser (QYQRES), and is capable of binding to PrP- Sc protein in fixed or unfixed tissue that has been treated to unmask the epitope to PrP-Sc protein and eliminate availability of a corresponding epitope of PrP-Cellular is described. A hybridoma cell line ATCC HB-12696 can produce and secrete the monoclonal antibody. Also described is a monoclonal antibody cocktail comprising the antibody that binds to the conserved epitope of prion proteins, designated as Gln-Tyr-Gln-Arg-Glu-Ser (QYQRES) and a second antibody that specifically binds to a second conserved epitope of prion proteins designated as Ile-His-Phe-Gly (IHFG). The continuous cell line ATCC HB-12403 can be produce the second antibody. The first antibody or the antibody cocktail can be used in the manufacture of an immunoassay for detecting the presence of PrP-Sc protein. |
