It is demonstrated that polycationic, non-lipid DNA formulations are resistant to nebulizer-induced degradation and are much more specific and efficient than lipid-based formulations in transfecting respiratory tract tissues in vivo. Results presented herein demonstrate that polyethyleneimine (PEI) formulations are 10-fold more effective than previous lipid-based formulations when delivered in vivo by jet nebulizer. Therefore, aerosolized polyethyleneimine-based formulations are provided herein for use in targeting gene therapy via the respiratory tract. |
