abstract | A bispidine derivative or a pharmaceutically acceptable derivative thereof has the formula (I) wherein: R1 and R2 independently are H, alkyl or together form -O-(CH2)2-O-, -(CH2)4- or -(CH2)5-; R9 is optionally substituted alkyl, phenyl, naphthyl, and alkylphenyl; R10 is H or -OH; R11 is H or alkyl; R12 is absent or is OH, cyano, halo, amino, nitro, alkyl, alkoxy, -NHS(O)2R16, -OS(O)2R16a, -NH-C(O)-NHR17 or -C(O)NHR18 ; A is a single bond, alkylene, -(CH2)nN(R20)-, -(CH2)nS(O)p-, -(CH2)nO- (wherein in three latter groups, the -(CH2)n group is attached to the carbon atom bearing R10 and R11), -C(O)N(R20)- (wherein in the latter group, the -C(O)- group is attached to the carbon atom bearing R10 and R11), -N(R20)C(O)O-(CH2)n-, -N(R20)-(CH2)n- (wherein in the two latter groups, the N(R20) group is attached to the carbon atom bearing R10 and R11) or -(CH2)mC(H)(OH)-(CH2)n- (wherein in the latter group, the -(CH2)m- group is attached to the carbon atom bearing R10 and R11); B is a single bond, alkylene, -N(R23)(CH2)r - or -O-(CH2)r- (wherein in the two latter groups, the -(CH2)r- group is attached to the bispidine nitrogen atom); m is 1, 2 or 3; n and r independently are 0, 1, 2, 3 or 4; p is 0, 1 or 2; q is 0; 1, 2 or 3; X is O or S; Cy1 is an optionally substituted five to seven-membered heterocyclic ring containing one or more heteroatom selected from O, N or S; R16, R16a, R20 and R23 independently are H or alkyl and R17, R18, R21 and R22 independently are H or alkyl; with the proviso that when the pharmaceutically acceptable derivative includes a N-oxide at the bispidine nitrogen, X is not S and/or p is not 0 when A is -(CH2)nS(O)p-. A pharmaceutical composition thereof is useful for the prophylaxis or treatment of arrhythmia. A compound of formula XVI wherein R10, R11, R12, A and B are as defined above or a protected derivative thereof provided that the compound is not 3-benzyl-9-oxo-3,7-diazabicyclo[3.31]nonane is disclosed. A compound of formula XVIII wherein R9 and X are as defined above is disclosed. |
