titleOfInvention | Indazole bioisostere replacement of catechol in therapeutically active compounds |
abstract | An indazole compound or a pharmaceutically acceptable salt thereof has the formula (I) wherein the compound and its therapeutic activity resulting directly from an indazole-for-catechol bioisostere replacement of a catechol containing compound having the therapeutic activity and the non-catechol substituents are the same or homologous before and after the replacement wherein: RC is independently hydrogen, hydroxy, -O-alkyl or optionally substituted phenyl; RA is independently hydrogen, -(CH2)n-cycloalkyl, or optionally substituted alkyl, alkylalkoxy, alkenyl, -(CH2)n-heterocyclyl or -(Z')b(Z")c-aryl; Z' is alkylene or alkenylene; Z" is -O-, -S-, -SO2- or -N(R9)-; n is 0 to 2; b and c are each independently 0 or 1; R9 is hydrogen or alkyl substituted by 0 to 3 fluorine atoms; RB is independently hydrogen, cycloalkyl or optionally substituted alkyl, alkenyl, phenyl, or alkyl-cycloalkyl Ra and Rb are independently hydrogen and non-catechol substituents of the compounds resulting directly from an indazole-for-catechol bioisostere replacement of the catechol-containing compound having the therapeutic activity, where the non-catechol substituents are the same or homologous before and after the replacement, provided that both of Ra and Rb cannot be hydrogen at the same time. A pharmaceutical composition thereof is useful for treating or preventing a disease or condition is mediated by or associated with interaction between receptors and ligands characteristic of the disease or condition, which are positively affected by the active ingredient. |
