abstract | Cephalosporins of formula (I) and their salts are new. In (I) R1 is H or protecting gp. R2 is organic gp. bonded via an O or S atom. esp. acetoxy or a substd. 5-(1,2,3,4-tetrazolyl)thio or 2-(1,2,3,4(sic)-thiadiazolyl)thio. R3 is lower alkyl. n is 0-2.A is H or opt. substd. alkyl. B is alkyl, cycloalkyl, cycloalkenyl, cycloalkadienyl or heterocyclyl, all opt. substd. They are prepd. e.g. by reacting a 7-(alpha-aminoacetylamino)cephem with a reactive deriv. of 4-A-2,3-dioxo-piperazine-1-carboxylic acid. (I) are esp. effective against Pseudomonas aeruginosa, Klebsiella pneumoniae and Proteus. They are useful in human or veterinary medicine. A typical cpd. 7 beta- D(-)-alpha-(4-ethyl-2,3-dioxo-1,piperazinecarboxamido)-alph- a-(2-thienyl)-acetamido -7 alpha-methoxy-3 5-(1-methyl,1,2,3,4-tetrazoyl)thiomethyl -DELTA3-- cephem-4-carboxylic acid (Ia) has intravenous LD50 in mice =5g/kg and has ID50 against beta-lactamase from E. coli GN-5482 of 0.82 mu g/ml. |
