abstract | Prepn. of penicillin or cephalosporin cpds. (I) is carried out by subjecting other penicillin or cephalosporin cpds. (II) to the action of an enzyme immobilised on a solid support. (II) are pref. used as 0.1-10% solns. The enzymes are pref. immobilised on CNBr-activated cellulose in the form of a powder, fibres, or a film. The cellulose is pref. preswollen before activation. The process can be used, e.g., to convert benzyl penicillin (IIa) to 6-aminopenicillanic acid (6-APA), to convert 6-APA to amoxycillin (Ia), or to convert 7-amino-desacetoxy- cephalosporanic acid (ADCA) to cephalexin (Ib). Use of immobilised enzymes yields high-purity products free of undesirable side-effects (e.g. allergenic activity). The immobilised enzyme can be readily recovered and re-used. |
