abstract | An alkali metal or ammonium insulin (I) is purified by adding to a column packed with a swollen gel having in the dry state a water regain of at least 4 wt. % and particle diameter smaller than 100 microns an aq. soln. of (I) of at least 80% purity, of concentration about 1-8% (w/v) and pH about 2. 0-3.5, to provide a column loading of about 0.8-6.7 g/1 of bed volume, and eluting the insulin from the column at about 5 degrees-30 degrees C with an aq. eluant having a pH in the same range as the insulin soln. by the present gel filtration, insulin having a higher purity than that obtained in similar prior art procedures is obtained. The product is usually subsequently converted to zinc insulin which may be made up into various pharmaceutical forms of insulin. Also the purer zinc insulin permits the prepn. of a neutral insulin which is more stable than acidic insulin; it also permits the formulation of extended action pork and beef insulin preparations without additional purification as was required in the past; further it permits the prepn. of isophane insulin using less protamine. |
