abstract | The disclosure provides an identification method of peptides binding with MHC I and their anchor residues by a random peptide library, including the following steps: (1) synthesizing random peptides with random sequence lengths of 8-12, (2) performing De Novo analysis on the random peptides, to obtain an amino acid distribution; (3) expressing the d chains and [32m chains of MHC I, (4) forming MHC I-peptide complexes from the random peptides, d chains and [32m chains in solution; (5) performing thermal denaturation, separation and purification on the MHC I-peptide complexes to obtain the peptides binding with MHC I, (6) performing De Novo analysis on the peptides binding with MHC I, to obtain an amino acid distribution of the peptides binding with MHC I, which was compared with the amino acid distribution in step (2) to give the major anchor positions and preferable anchor residues. The present method is simple to operate, has reliable results, takes short time, and achieves a high effective screening of potential pathogen-specific T cell epitopes, which could be extensively applied in preliminary screening of peptide vaccines for animals and human. |
