abstract | The invention provides compounds that have activity as inhibitors of cyclin-dependent kinases, glycogen synthase kinase-3 and Aurora kinases use in the treatment of disease conditions and conditions, such as ocmo cancer, which are mediated by kinases. The compounds have the general formula (I); where X is CR5 or N; A is a bond or - (CH2) m- (B) n-; B is C = O; NRg (C = O), or O (C = O), where Rg is hydrogen or hydrocarbyl of 1 to 4 carbon atoms optionally substituted by hydroxy or by alkoxy of 1 to 4 carbon atoms, m is 0, 1 or 2; n is 0 or 1; R0 is hydrogen or, together with NRg when present, forms a group - (CH2) p- where p is from 2 to 4; R1 is hydrogen, a carboxylic or heterocyclic group having from 3 to 12 ring members, or a hydrocarbyl group of 1 to 8 optionally substituted carbon atoms; R2 is hydrogen, a carboxylic or heterocyclic group having 3 to 12 ring members, or a hydrocarbyl group of 1 to 8 optionally substituted carbon atoms; R2 is hydrogen, halogen, methoxy or a hydrocarbyl group of 1 to 4 carbon atoms, optionally substituted by halogen, hydroxyl or methoxy; R3 and R4 together with the carbon atoms to which they are attached, form a molten carbocyclic or heterocyclic ring, optionally substituted, having 5 to 7 ring members of which up to 3 may be heteroatoms selected from N, O and S; and R5 is hydrogen, an R2 group or an R10 group wherein R10 is selected from halogen, hydroxy, trifluoromethyl, cyano, nitro, carboxy, amino, mono- or di-hydrocarbylamino of 1 to 4 carbon atoms, carbocyclic and heterocyclic groups which have from 3 to 12 ring members; a Ra-Rb group where Ra is a bond, O, CO, X1C (X2), C (X2) X1, X1C (X2) X1, S, SO, SO2, NRc, SO2NRc or NRcSO2; and Rb is selected from hydrogen, carbocyclic and heterocyclic groups having from 3 to 12 ring members, and wherein one or more carbon atoms of the hydrocarbyl group of 1 to 8 carbon atoms may optionally be replaced by O, S, SO, SO2, NRc, X1C (X2), C (X2) X1 or X1C (X2) X1; Rc is selected from hydrogen and hydrocarbyl from 1 to 4 carbon atoms; and X1 is O, S, or NRc and X2 is = O, = S, or = NRc. Also included in formula (I) are the salts, solvates and N-oxides of the compounds. |
