The invention relates to a method of producing a recombinant polyclonal protein composition, in particular a recombinant polyclonal antibody composition. The method comprises obtaining a collection of cells transfected with a library of variant nucleic acid sequences, wherein each cell in the collection is transfected with, and is capable of expressing a member of the library, which codes for a different member of a polyclonal protein that binds to a particular antigen and which is located in the same unique site in the genome of the individual cells in the collection, where the nucleic acid sequence is artificially related to the cell in the collection. The cells are grown under conditions suitable for expression of the polyclonal protein, which is obtained from the cells or the culture supernatant. The nucleic acid sequence is introduced into the cells by transfection with a vector library for site-specific integration. The present method is suitable for the manufacture of recombinant polyclonal antibodies and thus makes a superior substitution of plasma-derived immunoglobulin therapeutic products available. |
