abstract | Transgenes driven by naturally occurring cardiac promoters have relatively low levels of cardiac transgene gene expression, and consequently have limited the use of cardiac muscle as a target for plasmid-mediated gene supplementation. However, by randomly gathering motifs from elements of Caja E, MEF-2 TEF-1 and SRE, recombinant libraries of cardiac specific synthetic promoter have been produced. Examining the transcriptional activity both in vitro and in vivo of hundreds of resulting clones, a few specific cardiac synthetic promoters were discovered that contain a transcriptional power that greatly exceeds the transcriptional levels obtained from myogenic and viral natural gene promoters. These promoters are used to direct the expression of desirable genes in nucleic acid expression constructs specifically for cardiac cells. Thus, these cardiac specific synthetic promoters can be used for plasmid-mediated gene supplementation under serious health conditions, such as ischemic disease, myocardial infarction or heart attack. Thus, one aspect of the present invention is a cardiac specific synthetic promoter produced by a method that generates a library of random constructions of recombinant synthetic promoter expression. Another aspect of the present invention is directed to a method that uses the construction of cardiac specific synthetic expression for the expression of a gene of interest in a cardiac cell. |
