abstract Methods for introducing a polynucleotide into healthy tissue and generating a pulsed electrical field in the tissue by invasive electrodes are provided, resulting in improved supply of the polynucleotide into tissue cells, while minimizing local side effects for tissue. electroporated and systemic side effects for the electroporated organism due to metal contaminants released from the electrodes. In one embodiment, the methods of the invention use gold electrodes. Gold or other metal alloys that minimize the introduction of toxic amounts of metal into electroporated tissue. In other embodiments, the methods of the invention are used for gene therapy by administering DNA to cells of suitable target tissue, and for the induction of an immune response through the administration of a DNA vaccine.
