abstract | This invention relates to host cells, modified, novel, expressing heterologous, fused proteins, and methods for detecting test samples that have peptide binding activity; wherein the modified host cells comprise: (a) a genetic sequence encoding a heterologous fusion protein; the fusion protein consists of a first peptide of a peptide binding pair, or segment of the first peptide, which binds to a DNA binding domain, or the corresponding transcriptional activation domain of a protein for transcriptional activation; (b) a genetic sequence encoding a heterologous fusion protein, the fusion protein contains a second peptide of the peptide binding pair of (a), or a segment thereof, fused to a DNA binding domain or the domain of corresponding transcriptional activation, any of these is not used in (a); (c) a luciferessus gene operably associated with the transcriptional activation protein, or a part thereof; (d) as an option, a deletion or mutation in the chromosomal DNA of the host cell for the transcriptional activation protein, if present, in the selected host cell. |
