abstract |
A protein is described that is found in a natural way isolated from the saliva of the medicinal leech Hirudo medicinalis which is strongly bound to the collagen acting in this way as an inhibitor of the natural adhesion of platelets to the collagen. The protein has a molecular weight of approximately 12,000, an acidic isoelectric point and contains six cysteines. The protein is sequenced and the gene is cloned from an H. medicinalis cDNA library. Methods for producing such polypeptide by recombinant techniques are described. Recombinant and naturally occurring proteins are potent inhibitors of collagen-dependent platelet adhesion and therefore useful for the therapeutic treatment of various conditions related to heart disease and circulatory system diseases. Additionally, the protein is useful for coating the natural or artificial collagen surfaces in order to make them non-adhesive to the cells and prevent the activation of the cells. |
