abstract | High levels of serine-D take place in the mammalian brain, where it appears to be an endogenous binder of the "glycine site" of NMDA receptors. We have purified from a rat brain a soluble enzyme that catalyzes the direct racemization of serine-L to serine-D. The purified serine racemase has a molecular weight of 37 kDa and requires pyridoxal 5'-phosphate for its activity. The enzyme is highly selective towards L-serine, failing to racemize some other amino acid tested. We have also identified polynucleotide sequences that encode serine mamifera racemase, including human. Compounds that modulate the activity of the serine mammalian racemase are useful for treating conditions and diseases which involve overstimulation of NMDA receptors, such as heart attacks and various neurodegenerative diseases. |
