abstract | The present invention relates to a method for determining the percentage of glactose coated in a glycoprotein, which comprises: (a) preparing a glycoprotein in a medium for analysis, whereby a protein preparation is obtained; (b) preparing samples of enzymatically treated protein, and a control of the prepared protein comprising: dividing the protein preparation into a plurality of protein samples; (i) not adding enzyme to at least one protein sample as an antecedent control of medium sample, whereby sample A is obtained; (ii) add at least β-galactosidated to at least one protein sample, whereby sample B is obtained; (iii) add at least β-galactosidate and asialidase to at least one other protein sample, whereby sample C is obtained; and incubating the plurality of protein samples comprising samples A, B and C; (c) quantify the amount of galactose in each of samples A, B and C using HPAEC-PAD chromatography, where the amount of galactose in sample A reflects the amount of galactose in the medium, where the amount of galactose in sample B it reflects the amount of galactose not coated with a sialic acid residue in the protein preparation in addition to the amount of galactose in the medium, and where the amount of galactose in sample C reflects the amount of galactose coated with a sialic acid in addition to the amount of galactose in the medium and not coated by sialic acid; and (d) determine the percentage of galactose coated in the glycoprotein based on the amounts of galactose in sample A, sample B and sample C, wherein the method has the linearity of R2 = 0.99. |
