abstract | Process for the preparation of the compounds of general formula I, among them, misoprostol (see Formulas) where R represents a C1-4 alkyl group of straight or branched chain by coupling with cuprate of the vinyl cuprate of general formula II prepared by reacting the vinyl stannane of general formula III with copper halide CuX and alkyllithium R1Li (see Formula) where: R2 represents H or an alcohol protecting group that may contain a silicon atom, such as, for example, a trimethylsilyl, triethylsilyl, tert -butyldimethylsilyl, or a cyclic or open chain alkyl group containing an oxygen atom, such as a tetrahydropyranyl, methoxymethyl or ethoxymethyl group; X means I, Br, CN group, SCN, OSO2CF3, R1 represents a C1-6 alkyl group, n>2, if R2 is not a hydrogen atom, n>3, if R2 is a hydrogen atom, and the enone of the general formula IV (see Formula) where R3 represents a THP or trialkylsilyl group and the meaning of R is as defined above takes part in the cuprate reaction in such a way that a.) the excess of the alkyllithium, which is applies in comparison with Cu(I) iodide. in the case of R2 ? H in a molar ratio of 2 to 2.4, in the case of R2 = H in a molar ratio of 3 to 3.4, decomposes before the coupling reaction of II and IV, b.) the protecting groups of the resulting compound of the general formula V (see Formula) where the meanings of R, R2 and R3 are as defined above, are eliminated, the obtained compound of general formula I is purified by chromatography. |
