abstract |
A method of selecting cells having zero level of fucose, useful as host cells for the expression of recombinant proteins, is described; The method comprises: (d) introducing genetic mutations into a population of CHO cells by contacting the cells with methotrexate (MTX), (e) contacting the population of CHO cells comprising cells mutated with an agent that binds to the non-toxic fucose for an amount of time that allows the binding of the agent that binds to the fucose to a portion of fucose on the membrane of a cell in the cell population, where the amount of time does not allow the destruction of cells ; and (f) depleting from the population of cells comprising mutated cells, a subpopulation of cells that bind to the agent that binds to the fucose, thereby selecting useful cells as host cells for the expression of recombinant proteins, the selected cells having zero fucose content; Cells and cell lines useful as host cells for the expression of recombinant proteins are also described. |
