abstract | A conjugate is presented between an antithrombotic agent and a bioabsorbable polymer. In addition, a method is presented for applying a coating comprising a bioabsorbable polymer conjugate and an antithrombotic agent to at least a portion of an implantable device to prevent or reduce thrombosis formation on the surface of the device. A first layer or sublayer of the coating is prepared by mixing a polymeric material and a biologically active agent with a solvent, thus forming a homogeneous solution. The second outer layer or layer comprises a bioabsorbable heparin-polymer antithrombotic conjugate. This coating is applied to the inner layers containing the drug using, for example, immersion or spray coating processes. After drying, the bioabsorbable heparin polymer antithrombotic conjugate remains in the outer layer of the coating, allowing the agent in the inner layer to elute through it. Also, the outer layer prevents thrombosis, and can modulate the release kinetics of the agent (s) that are contained in the inner layer (s) of the coating. |
