abstract The present invention relates to methods of generating conjugated immunoglobulins, the method comprising: stripping a cysteine at amino acid position 80 ("cys80") in a light chain variable region of an immunoglobulin, immunoglobulin comprising a heavy chain variable region and the light chain variable region; and conjugation of a thiol reactive compound with cys80, the thiol reactive compound comprising a thiol reactive group. The invention further relates to antigen binding molecules and methods of generating same, immunoglobulins as well as nucleic acid molecules encoding the immunoglobulins and host cells comprising the nucleic acid molecules, immunoglobulins. conjugated immunoglobulins, and light chain variable regions for use in conjugated immunoglobulin.
