abstract |
This invention provides novel intermediates and derivatives of 1,4-benzothiazepine, and methods for their synthesis and assay. It also provides methods of using these novel compounds to limit or prevent a decrease in the level of FKBP12.6-RyR2 complex in a subject; to prevent sudden cardiac death during physical exercise; treat or prevent heart failure, atrial fibrillation or cardiac arrhythmia produced by exercise. The present invention further provides methods of detecting the agent that enhances the binding of RyR2 and FKBP12.6, and the agents detected by these methods. In addition, this invention provides a method of detecting agents for the treatment and prevention of cardiac failure, atrial fibrillation or arrhythmia produced by exercise, and prevention of cardiac death subtelors of exercise. It also provides the agents identified by such methods. |
