abstract | The present invention provides an animal tissue clearing method, and belongs to the technical field of animal tissue observation. The method includes the following steps: (1) anesthetizing an animal, sequentially perfusing heart perfusate and a tissue fixation solution to a heart of the animal, then dissecting the animal, and separating animal tissue; (2) placing bone tissue in a decalcification solution, and performing decalcification for 5-12 d to obtain decalcified bone tissue, where solution change is conducted once every 1-3 days in the decalcification process; (3) respectively placing decalcified bone tissue and soft tissue in a destaining solution, and performing destaining to obtain destained tissue; (4) placing the destained tissue in a degreasing solution, performing gradient degreasing, placing degreased tissue in a dehydration solution, and performing dehydration to obtain dehydrated tissue; and (5) placing the dehydrated tissue in a clearing solution, and performing clearing treatment so as to obtain clear tissue. |
