A method of adding adapters to fragmented nucleic acids for next-generation sequencing, comprising providing multiple codes based on varying adapter molecule barcode lengths on both sides of fragmented nucleic acids, and from the same fragment based on both barcodes. A method is disclosed that includes identifying a read. The methods and articles enable amplification of fragmented nucleic acids in the presence of low yields of isolated fragmented nucleic acids, and also enable efficient and reliable detection of low frequency mutations, including subpopulations of cells in a subject. do. |
