abstract | Certain exemplary embodiments of the present invention are A method for isolating and concentrating nucleic acids of a selected target size (eg, less than 1000 base pairs increments) from a biological fluid mixture, the biological fluid mixture comprising: a first phase solution in which a first phase forming polymer or surfactant component is dissolved; and a first aqueous two-phase system (ATPS) formed from a second phase solution such that target nucleic acid fragments smaller than a desired target size are partitioned into the second phase solution and contaminants are partitioned into the first phase solution. ; The second phase solution is extracted and mixed with a second ATPS formed of a third phase solution and a fourth phase solution in which the second phase forming polymer or surfactant component is dissolved to distribute and concentrate the target nucleic acid fragment into the third phase solution to become; and recovering the concentrated target nucleic acid fragment from the third phase solution. The present invention also includes compositions and kits for isolating and enriching nucleic acids of a selected target size as described above according to certain exemplary embodiments. |
