abstract | The complement system is part of the innate immune system and is highly regulated by regulatory proteins belonging to the complement activation regulator (RCA) family. Lack of regulation causes damage to host cells, and deficits in regulation are also known to be associated with diseases such as, for example, age-related macular degeneration, atypical hemolytic uremic syndrome, and dense deposit disease. Regulatory proteins mainly include decay promoting factor (DAF; CD55), membrane cofactor protein (MCP; CD46), complement receptor 1 (CR1; CD35), factor H (FH) and C4b-binding protein (C4BP). Structurally, these proteins are composed of repeating complement control protein (CCP) domains in which 2-4 contiguous domains contribute to regulatory functions of decay-promoting activity (DAA) and cofactor activity (CFA). However, there are currently no four domain chimeric proteins with both potent DAA (CP-DAA and AP-DAA) and potent CFA (C3b-CFA and C4b-CFA). Thus, the chimeric protein DCP (ie, dual activity regulator) was generated. |
