abstract |
Twin Immune Cell Engager (TWICE) is a kit or composition for treating cancer comprising a first component and a second component. Each component comprises a targeting moiety that binds to a tumor antigen expressed by the cancer or an antigen expressed by non-cancer cells in the tumor microenvironment. In some embodiments, the first and second components are each complementary to an immune cell binding domain capable of immune cell binding activity when bound to the immune cell binding domain of the other component, and complementary to the complementary binding domain of the other component. and a complementary binding domain capable of binding an antigen. In some embodiments, the first and/or second component comprises a complementary functional domain that has activity when targeted to a cancer cell or its microenvironment. The present TWICE complex provides a unique ability to combine multiple functions into a two-component complex that is activated in the tumor microenvironment. Thus, the present complex provides a significant advantage in having a single approach to administering a two-component complex that is localized to the tumor microenvironment and can result in two different signals of benefit to the patient. This unique construct provides advantages not present in prior art constructs. |
