The present invention generally relates to at least a first cleavable moiety (CM1) that is a substrate for at least one matrix metalloprotein (MMP) and at least one serine protease (SP) or at least one cysteine protease a polypeptide comprising at least a second cleavable moiety (CM2) that is a substrate for (CP), an activatable antibody and at least CM1 that is a substrate for at least one MMP protease and at least one SP protease or at least other larger molecules comprising such a polypeptide comprising at least CM2, which is a substrate for one cysteine protease (CP), and for at least one MMP protease in a variety of therapeutic, diagnostic and prophylactic indications. Methods of making and using such polypeptides comprising at least CM1 as a substrate and at least CM2 as a substrate for at least one SP protease or at least one cysteine protease (CP) are provided. |
