abstract The present invention relates to a method for isolating and purifying anti-inflammatory fat A and core O-antigen that promotes immune cell activity from lipopolysaccharide of a Salmonella typhimurium mutant strain with weakened virulence. In the present invention, lipopolysaccharide is safely extracted from cultured cells of a mutant strain with weakened virulence that synthesizes the same lipopolysaccharide as that of Salmonella Typhimurium strain, and 0.25% trifluoroacetic acid solution Fat A and core O-antigen polysaccharides were hydrolyzed by incubation at 50 °C, and fat A and core O-antigen polysaccharides were separated with a Sep-Pak tC18 cartridge, and the gel filtration conditions were optimized to reduce the endotoxin effect. A size exclusion chromatography method for purifying single fractions of fat A and core O-antigen polysaccharide with high efficiency and high purity is provided. And a method for inhibiting the inflammatory response of mouse macrophages and promoting immune cell activity using the purified fat A and core O-antigen polysaccharide will be described.
