titleOfInvention | Precisely engineered insidious messenger RNA and other polynucleotides |
abstract | The present disclosure relates to methods of lowering immunogenicity in long polynucleotide sequences by precisely sequencing immunogenic motifs within the polynucleotide sequence. The present disclosure further relates to precisely sequence engineered polynucleotides with improved functionality, such as exhibiting low innate immunogenicity, improved stability or high protein expression. In these polynucleotides, the immunogenic sequence motif is removed, while the remainder of the sequence is conserved. This targeted engineering approach to total nucleotide changes has unique advantages, which include less disruption of natural or optimized polynucleotide sequences and, moreover, high expression retention, but possible stealthiness to innate immune receptors. including that |
