abstract Methods of characterizing genome editing, clonal expansion, and associated reagents for use in such methods are disclosed herein. Some embodiments of the present disclosure, in some embodiments after an engineered genome editing event, comprising characterizing genomic alterations that occur at both the intended and unintended genomic loci within the genome of the cell population. It's about identifying cell populations. Another embodiment relates to the use of duplex sequencing to assess clone selection in a mixed cell population and/or cell population after a genome editing event. A further example of the present disclosure relates to methods of detecting and evaluating clonal expansion of cells after a genome editing event.
