The present invention provides methods for genetically engineering T cells, such as CD4+ T cells and/or CD8+ T cells, for use in cell therapy. In some aspects, provided methods include, for example, pooling concentrated CD4+ and CD8+ cells in a ratio of 1:1, followed by incubating the cells under stimulation conditions, and transduction or transfection. Introducing the recombinant polypeptide into the cell through (transfection) and/or cultivating the cell under conditions that promote proliferation and/or amplification. In some aspects, the provided method is an efficient and reliable means of generating genetically engineered T cells with high success. |
