Methods and compounds for increasing sensitivity of botulinum assays |
abstract |
The device, system, and method may provide for improved detection of botulinum neurotoxin. In one aspect, isoquinolinyl compounds can be used to enhance the sensitivity of both FRET and non-FRET cell-based assays. In another aspect, non-FRET assays and constructs use reporters that are not linked in a manner that produces significant FRET with the second fluorescent material. In this topic, environmental cells may contain enzymes that significantly degrade reporter degradation after cleavage than before cleavage, and the presence of botulinum toxin is associated with signal reduction from the reference signal. If the environment is a cell, the cell may preferably express both a construct comprising a reporter and an enzyme that enables degradation. |
