The present invention is directed to a method of modifying a peptide composition to increase stability and delivery efficiency. In particular, the present invention relates to methods of increasing the stability and delivery efficiency of protein kinase C (PKC) regulatory peptide compositions. "Therapeutic peptide composition" includes "carrier peptide" and "cargo peptide". A "carrier peptide" is an amino acid sequence within a peptide or peptides that promotes cellular uptake of the therapeutic peptide composition. "Cargo peptides" are PKC regulatory peptides. As described herein, peptide modifications to carrier peptides, cargo peptides, or both, can result in stability and delivery efficiency of therapeutic peptide compositions by reducing disulfide bond exchange, physical stability, reduction of proteolytic degradation, and increased cell uptake efficiency.
