In the present invention, a method for purifying disintegrin-like and metalloproteinase (ADAMTS13) recombinant protein with thrombospondin type 1 motif 13 from a sample is provided. The method comprises concentrating the ADAMTS13 protein by chromatographically contacting the sample with hydroxyapatite under conditions such that the ADAMTS13 protein can appear in the eluate or supernatant from the hydroxyapatite. These methods may further comprise tandem chromatography with mixed modal cation exchange / hydrophobic interaction resins that bind to ADAMTS13 protein. Additional optional steps involve ultrafiltration / diafiltration, anion exchange chromatography, cation exchange chromatography, and virus inactivation. In addition, the present invention provides a method of inactivating viral contaminants in a protein sample, wherein the protein is immobilized on a support. Also provided herein are compositions of ADAMTS13 prepared according to these methods. |
