abstract | Two-chain insulin analogues include modified A-chain polypeptides and modified B-chain polypeptides. The modified A-chain polypeptide may comprise a His or Glu substituent at the A8 position; Glu substituent at the A14 position; And one or more Gln or Arg substituents at the A17 position. The modified B-chain polypeptide may be selected from the group consisting of a deletion of amino acids at the B1, B1-B2, B1-B3, B30, or a combination thereof; Ala or Glu substituent at the B2 position; And one or more Glu substituents at the B3 position. The analogs show thermodynamic stability, reduced magnetic binding, maintenance of biological potency, and increased mitosis in zinc-free solutions. The analogs show resistance to chemical and physical degradation. A method for treating a diabetic or obese patient includes administering to a patient a physiologically effective amount of an insulin analog or a physiologically acceptable salt thereof. |
