abstract |
A composition, method and system for determining the sequence of a template nucleic acid using a sequencing procedure by polymerase based binding are provided. The testing step comprises monitoring the interaction between the polymerase and the template nucleic acid in the presence of one or more nucleotides. The following exact nucleotide entity in the sequence is determined without the introduction of any nucleotide into the structure of the primer by the formation of a phosphodiester linkage. An optional introduction step can be used after the inspecting step to extend the primer by one or more nucleotides, thereby increasing the template nucleotides that can be examined in subsequent testing steps. The sequencing procedure by conjugation does not require the use of labeled nucleotides or polymerases, but optionally these reagents can be used. |
