Methods and compositions for transfecting cells with a plasmid are described. In certain embodiments, methods and compositions are disclosed wherein the efficiency of transfection is significantly increased by contacting the nucleic acid-free polyethyleneimine (PEI) with the cells to be transfected during the transfection process. Therapeutically useful adeno-associated viral vectors produced according to the described methods and compositions are also described. |
