abstract |
The present invention relates to gag and pol proteins; An envelope protein or functional replacement thereof; And a retrovirus producing cell comprising a nucleic acid sequence encoding an RNA genome of a retroviral vector particle, wherein the nucleic acid sequence is all located in a single locus in the retrovirus producing cell genome. The present invention also encompasses methods and compositions that include a retroviral nucleic acid sequence that encodes a non-mammalian replication origin and the ability to retain DNA of at least 25 kilobases (kb) and that encodes gag and pol proteins, and env proteins or functional alternatives thereto ≪ / RTI > The present invention also relates to methods and uses of said nucleic acid vectors to produce stable retroviral packaging and producing cell lines. |
