Compositions and methods relating to ApoA-1 fusion polypeptides are disclosed. The fusion polypeptide comprises a first polypeptide segment corresponding to an ApoA-I polypeptide or an ApoA-I mimic, and may also comprise a dimerization domain, e. G., An Fc region, typically through a flexible linker Terminal to the first polypeptide segment. In some embodiments, the fusion polypeptide further comprises a second polypeptide segment located at the carboxyl-terminus for the first polypeptide segment, which is further characterized by a second biological activity (e. G., RNase, paraoxonase, platelet- Acetyl hydrolase, cholesterol ester transfer protein, lecithin-cholesterol acyltransferase, or a polypeptide specifically binding to amyloid beta). Also disclosed are dimeric proteins comprising first and second ApoA-I fusion polypeptides as disclosed herein. Fusion polypeptides and dimeric proteins are useful in methods for therapy. |
