abstract The present invention is directed to defined multijunction oligonucleotides that can have a predetermined size and composition. For example, in various embodiments, the defined multijunction oligonucleotides are in the form of defined multijunction siRNAs having an advantageous property, such as enhanced intracellular delivery and / or multi-gene suppression effects (i. E., 2,3 &Lt; / RTI &gt; or more siRNA). In various embodiments, defined multidentate oligonucleotides can be synthesized by the novel synthetic intermediates and methods. Defined multidentate oligonucleotides can be used, for example, to reduce gene expression, to study biological studies, to treat or prevent medical conditions, or to generate novel or modified phenotypes in cells or organisms.
