abstract The present invention provides a method for quantifying MSLN protein directly in a biological sample, comprising a sample immobilized to formalin by selective reaction monitoring / multiple reaction monitoring (SRM / MRM) mass spectrometry. Samples that can be analyzed include, but are not limited to, formalin fixed cells / cells, formalin fixed / paraffin embedded (FFPE) tissue / cells, FFPE tissue blocks and cells from this block, and formalin fixed and / Containing tissues and cells treated with formaldehyde-containing formulation / fixative. A method for producing a protein sample from such a biological sample is provided, wherein the MSLN protein is quantitated by SRM / MRM mass spectrometry by quantifying at least one of the peptides described in the protein sample.
