abstract |
Methods and preparations for preparing concentrated mitochondrial preparations using reverse phase centrifugation are provided. The method includes centrifuging a closed bottom tube containing a heavy phase in the proximal tube section relative to the rotational axis and a light phase in the distal tube section relative to the rotational axis. Centrifugation causes the suspension to form a thickened mitochondrial layer at the interface between the heavy and hard layers, and collects the fluid at this interface to produce a concentrated mitochondrial formulation. Concentrated mitochondrial preparations can be used in a variety of applications, including in vitro fertilization. |
