abstract | Amino acid residue incorporation is essentially found in low concentration sequence variants mixed with the expressed polypeptide, resulting from one or more base mismatches in codons that are susceptible to amino acid residue incorporation during transcription and / or translation. The present invention provides a method for optimizing the coding sequence of a polynucleotide encoding a polypeptide wherein at least one codon is susceptible to incorporation of amino acid residues. The methods of the present invention can be used to reverse engineer unknown coding sequences that differ from coding sequences known in the art, coding for the same polypeptide but at least one of the above codons. In addition, such methods can be used to alter the immunogenic potential of the expressed polypeptide. Thus, the present invention is useful for engineering optimized polynucleotides encoding polypeptides. |
