abstract Spherical nucleic acids (SNA), consisting of dense packed and highly oriented polynucleotide strands attached to nanoparticle surfaces, can overcome difficulties in conventional nucleic acid delivery. The present disclosure demonstrates that G-enhanced SNAs show uptake into cells several times larger than SNAs in which the other nucleotides are increased. This disclosure is applicable to other nanoparticle systems and provides an effective strategy for maximizing the intracellular delivery of SNAs, which establishes important design considerations for nanoparticle-based intracellular delivery of therapeutic agents.
