abstract In one aspect, the present application provides an Fc-containing polypeptide conjugate comprising an Fc-containing polypeptide conjugated to a conjugate moiety, wherein the Fc-containing polypeptide comprises a receptor glutamine residue located next to the N-glycosylation site Glycosylated Fc region, wherein the conjugate moiety is conjugated to the Fc-containing polypeptide via a receptor glutamine residue. Also provided are methods of producing such Fc-containing polypeptide conjugates using wild-type or engineered transglutaminase. Also provided is a manipulated transglutaminase system specifically designed to perform this reaction.
