The present invention provides a compound of formula I wherein R 12 or R 13 is a phosphonate (R 32 ), or a pharmaceutically acceptable salt or composition thereof, wherein said compound is an inhibitor of complement factor D, And a manufacturing method thereof. The inhibitors described herein target factor D and inhibit or modulate complement cascade at early and critical points in other complement pathways and reduce the ability of factor D to modulate the typical lectin complement pathway. Inhibitors of factor D described herein can reduce certain autoimmune, inflammatory and neurodegenerative diseases, and excessive activation of complement that is associated with ischemic reperfusion injury and cancer. |
